dc.description.abstractRESUMEN La mala gestión de los residuos por parte de determinados sectores productivos, como el industrial o el agrícola, ha propiciado un incremento de la acumulación de compuestos orgánicos persistentes (COPs) de origen sintético en la naturaleza a lo largo del último siglo, poniendo en peligro la seguridad humana y de los ecosistemas debido a su carácter altamente tóxico y contaminante. Dentro de este amplio grupo de compuestos cabe destacar, por su facilidad para evaporarse a temperatura ambiente, los conocidos como Compuestos Orgánicos Volátiles (COVs), entre los que se halla el acetonitrilo, un disolvente muy empleado a nivel industrial y a escala de laboratorio, que puede producir graves efectos medioambientales. En consecuencia, urge el desarrollo de técnicas sostenibles para su tratamiento, entre las cuales destaca la biorremediación por su demostrada fiabilidad y rendimiento económico. Con el fin de encontrar microorganismos con alto potencial para ser empleados en tareas de biorremediación de contaminantes orgánicos, concretamente, de acetonitrilos, se evaluó la capacidad degradadora de una colección de 16 cepas del género Rhodococcus. Para ello, en primer lugar, se probó su tolerancia al potencial tóxico del acetonitrilo y, seguidamente, su capacidad para crecer utilizando el acetonitrilo en procesos de cometabolismo o como única fuente de carbono y nitrógeno. Tras seleccionar las mejores cepas, se cuantificó la capacidad de éstas para metabolizar el acetonitrilo. Por último, se comprobó in vitro la capacidad protectora de las cepas mediante un bioensayo de fitotoxicidad sobre semillas y, tras seleccionar finalmente la cepa más prometedora de la colección, se llevó a cabo la búsqueda molecular de genes asociados a la degradación de nitrilos. De las 16 cepas de Rhodococcus que constituyeron la colección de estudio del presente TFG, fue la cepa 487, identificada como Rhodococcus rhodochrous, la que mostró los mejores resultados para el conjunto de ensayos, convirtiéndola en la mejor candidata para tareas de biorremediación de nitrilos. ABSTRACT The inadequate waste management carried out by certain productive sectors, such as the industrial or agricultural, has caused an increase in the accumulation of synthetic persistent organic pollutant (POP) compounds in nature over the last century, jeopardizing human and ecosystems security due to its highly toxic and polluting character. Within this wide group of compounds, it is worth noting those known as Volatile Organic Compounds (VOCs) due to its ease of evaporating at room temperature, among which is acetonitrile, a solvent widely used in industry and laboratories, that can produce serious environmental effects. In consequence, it is urgent to develop new strategies for its sustainable treatment, such as bioremediation, which has proven to be highly reliable and cost-effective. With the aim to find microorganisms with high potential in the bioremediation of organic pollutants, specifically acetonitriles, the degrading capabilities of 16 strains of the Rhodococcus genus were evaluated. To that end, firstly, its tolerance to the toxic potential of acetonitrile was tested, and then, their capacity to grow using acetonitrile in co-metabolic processes or as the single carbon and nitrogen sources available. After selecting the best strains, their potential to degrade acetonitrile was measured. Finally, the protective capacity of the strains was verified in vitro through a phytotoxicity bioassay on seeds and, after selecting the most promising strain in the collection, the molecular search for genes associated with nitrile degradation was carried out. Of the 16 Rhodococcus strains that comprise the collection shown in this Bachelor Thesis, it was strain number 487, identified as Rhodococcus rhodochrous, the one that showed the best results for the set of tests, turning it to be the best candidate for nitrile bioremediation tasks.es_ES
